Can I mount a suppressor to the threaded barrel without removing the compensator?

No. The compensator threads onto the barrel using taper screws and must be removed to expose the 1/2×28 threads for a suppressor. Removal is straightforward with the included hex wrench.

Will this fit standard P320 holsters?

Because of the compensator and threaded barrel, the Spectre Comp is longer than a standard P320 Compact. Many P320 Full-size holsters will work, but you should verify that the holster is open-ended or designed for a threaded barrel.
